摘  要:目的观察低氧习服对模拟高原低氧大鼠肺组织的影响及其HIF-1α表达变化。方法56只Wistar大鼠随机分成7组,每组8只:常氧对照组(N组)、急性低氧对照组(H0组)、急性低氧组(H1组)、3000m低氧习服组(C3.0组)、3000m低氧习服+急性低氧组(C3.1组)、5000m低氧习服组(C5.0组)、5000m低氧习服+急性低氧组(C5.1组n=8)。习服完成后,置于模拟海拔6000 m急性低氧24小时,检测动物习服完成后肺组织HIF-1α表达情况及不同低氧习服水平对大鼠急性低氧后肺结构改变的影响。结果急性低氧组肺组织显微和超微结构出现明显的间质性肺水肿,而经过低氧习服后间质性肺水肿则明显减轻;Western Blot方法未检测到各习服组肺组织表达HIF-1α,但HIF-1α mRNA在N组有一定量的表达,而C3.0组和C5.0组HIF-1α mRNA表达均明显高于对照组。结论低氧习服后肺组织HIF-1α mRNA表达的提高有利于减轻大鼠低氧性肺水肿。Objective To investigate effects of hypoxic acclimatization on HIF-1α expression in rats and on the pulmonary structure in rats. Methods Fifty-six Wistar rats were randomly divided into 7 groups, including normal control group (N, n=8), acute hypoxic control group (H0, n=8), acute hypoxic group (H1, n=8), 3000m hypoxic acclimatization group (C3.0, n=8), 3000m hypoxic acclimatization complicated acute hypoxic group (C3.1, n=8), 5000m hypoxic acclimatization group (C5.0, n= 8), 5000m hypoxic acclimatization complicated acute hypoxic group (C5. 1, n= 8). After treated with hypoxic acclimatization, animals were exposed to a simulated altitude of 6000m for 24 hours. Then, we assessed the expression of HIF-1α in the lungs and observed the structure of the lungs. Results Pulmonary interstitial edema was apparent in H1 group by light microscope and electron microscope, while significantly relieved in the two hypoxic acclimatization groups, HIF-1α protein could not be detected in all groups of rat lungs with western blot. Compared with that in N group, the levels of HIF-1α mRNA significantly increased in C3.0 group and C5.0 group (P〈0. 01). Conclusions Hypoxie acclimatization might improve the hypoxie pulmonary edema through increasing HIF- 1α in rats.
