摘  要:目的:探讨14-3-3ζ对肺癌细胞转移的押制作用。方法:以人肺巨细胞癌低转移细胞株为模型,利用RNAi技术研究14-3-3ζ被干涉后肿瘤细胞恶性表型的变化,探讨14-3-3ζ与肿瘤转移的相关性。结果:本研究发现14-3-3ζ被干涉后细胞的集落形成能力明显提高,黏附能力明显降低,细胞迁移、侵袭能力明显提高。结论:14-3-3ζ可能抑制肺癌细胞的转移。Objective:To investigate the inhibition of metastasis of lung cancer cells by 14-3-3ζ. Methods: RNAi technology was employed. The RNAi expression vectors of 14-3-3ζ were constructed and transfected into PLA-801C (the poorly metastatic subline of human lung giant cell carcinoma) . The cell cycle alteration, cell's ability of proliferation, adhesion, migration and invasion after transfection were studied. Results: Cells transfected with RNAi vector showed much lower 14- 3-3ζ expression and ability of adhesion, but much better ability of proliferation,migration and invasion. Conclusion: 14-3- 3ζ may inhibit the metastasis of lung cancer cells.
