Pg-LPS对束缚应激大鼠腹腔巨噬细胞凋亡的影响

摘　　要：目的:检测牙龈卟啉菌内毒素(Pg-LPS)对束缚应激大鼠腹腔巨噬细胞凋亡情况的影响,初步探讨应激影响牙周疾病的机制。方法:采用束缚应激方式,将大鼠随机分为正常对照组、应激组,于应激结束时处死动物,常规收集腹腔液。巨噬细胞于贴壁纯化后分别用RPMI-1640培养液,RPMI-1640培养液+0.01μg/mlPg-LPS,RPMI-1640培养液+1μg/mlPg-LPS继续培养。于24h后收集细胞爬片,采用荧光染色法、原位末端标记法(TUNEL)观察巨噬细胞凋亡情况。结果:1μg/mlPg-LPS刺激后,正常对照组及应激组巨噬细胞的凋亡率明显增加,应激组巨噬细胞凋亡率显著高于正常对照组。结论:一定浓度的Pg-LPS刺激后,大鼠腹腔巨噬细胞发生凋亡;束缚应激能加重这种异常。Objective：To explore the effects of restrain stress on the apoptosis of Pg-LPS stimulated peritoneal macrophages in Rats. Method： Rats （SD, male） were tested by using restrain stress. Rats were divided into 2 experimental groups randomly （control and stress）.After stress conditions, the rats were sacrificed, and their peritoneal macrophages were isolated and cultured. Cells were stimulated with 1μβg/ ml and 0.01 μg / ml Pg-LPS. After 24 hours, cells were collected and observed with fluorescence staining and TUNEL. Result：The apoptosis rate of macrophages in the stress and control group increased after 1 μg/ml Pg-LPS stimulate. Compared with the control,the apoptosis rate of macrophages in the stress group is significantly higher. Conclusion： The results suggest that chronic restrain stress modulates the apoptosis of Pg-LPS stimulated macrophages.
