摘 要:目的:调查广西省南宁市上尧码头是否存在莱姆病疫源地。方法:用BSK-Ⅱ培养基培养的方法从捕获鼠的膀胱分离莱姆病螺旋体。用PCR扩增分离菌株的5S-23S rRNA基因间隔区,将PCR扩增产物纯化回收后测序,通过将测序结果与基因库中莱姆病螺旋体菌株的5S-23S rRNA基因间隔区进行同源性比较和分析,从而鉴定分离株的基因型。结果:先后从上尧码头捕获的鼠的膀胱分离到了3株疏螺旋体,分别命名为GXSH4、GXSH7及GXSH9。GXSH7株经鉴定为B.valaisiana基因型莱姆病螺旋体。结论:南宁市上尧码头存在莱姆病疫源地。Objective: To explore the natural loci of Lyme disease in Shangyao port of Nanning of Guangxi province. Methods: We isolated Borrelia burgdorferi sensu lato which was the causative agent of Lyme disease from rats' urinary bladder with BSK - Ⅱ medium. PCR technique was used to amplify the 5S -23S ribosomal RNA intergenic spacer of the isolated strain. The purified positive PCR products were sequenced and analyzed to define the genospecies of the isolate. Results:Three strains of spirochete, which were named GXSH4, GXSH7 and GXSH9 respectively, were isolated from the collected rats. The strain of GXSH7 was identified by analysis of restriction polymorphism of ribosomal 5S -23S spacer amplicons. The result showed that it was more related to those of Borrelia burgdorferi, and fell into a cluster representing B. valaisiana species in the phylogenetic tree. Conclusion: Shangyao port of Nanning exists the natural loci of Lyme disease.
