摘 要:目的观察骨髓间充质干细胞(MSC)对肝癌细胞增殖能力的影响。方法人肝癌细胞系MHCC97-H培养液中分别加入0.25%和50%MSC的条件培养基(MSC-CM),采用CyQUANT细胞增殖实验检测吸光度A值变化。在MHCC97-H细胞培养液中添加50%MSC-CM,荧光实时定量聚合酶链反应(RT-PCR)检测肿瘤增殖细胞核抗原(PCNA)、Ki-67抗原的变化。16只裸鼠皮下接种MHCC97-H细胞后,实验组经静脉注射MSC,1×10。个/次,每周3次,对照组静脉注射PBS;比较肿瘤体积。结果培养液中加入MSC-CM后A值依次为211.65±54.72、236.24±57.15和283.59±62.16(P〈0.05)。PCNA和Ki-67的表达水平分别升高1.8和7.0倍。实验组肿瘤体积(1745±455)mm^3大于对照组(972±568)mm^3(P〈0.05),肿瘤体积平均增加速度(56.0±26.1)mm^3/d高于对照组(32.4±14.5)mm3/d(P〈0.05)。结论骨髓间充质干细胞可促进肝癌细胞系MHCC97-H的增殖。Objective To investigate the possible role of mesenchymal stem cells (MSC) in the proliferation of hepatocellular carcinoma (HCC) cells. Methods The HCC cell line (MHCC-97H) was incubated with 0, 25% and 50% MSC condition medium ( MSC-CM ) respectively and tested by CyQUANT cell proliferation assay. After MHCC97-H cells were cultured with 50% MSC-CM, the expression levels of proliferating cell nuclear antigen (PCNA) and Ki67 were tested by Real time-PCR. Sixteen nude mice inoculated subcutaneously with MHCC97-H cells were treated with 1 × 10^6 MSC or PBS via in- travenous injection three times per week. The tumor sizes were evaluated. Results After MHCC97-H cells were treated with 0,25% and 50% MSC-CM, A values were 211.65 ± 54.72,236.24 ± 57. 15 and 283.59 ± 62.16 respectively (P 〈0.05 ). PCNA and Ki-67 expression levels in MHCC97-H cells treated with MSC-CM were increased by 1.8 and 7 times, respectively. The tumor volume in MSC-treated group (1745±455) mm^3 was much bigger than that in the controls (972 ±568) mm^3 ,P 〈0.05,and also the average tumor growth rate (56.0 ±26.1 ) mm^3/day was significantly increased as compared with the controls (32.4 ± 14.5 ) mm^3/day,P 〈 0.05. Conclusion MSC can promote both in vitro proliferation and in vivo tumor growth of HCCs.
