摘  要:目的探讨巨噬细胞游走抑制因子(MIF)在结肠癌肿瘤组织和细胞系中的表达,探讨其表达和肿瘤血管发生的关系。方法免疫组织化学法检测人结肠癌组织中MIF表达,同时用CD34单克隆抗体标记结肠癌组织切片中新生血管,计算微血管密度(MVD),酶联免疫吸附分析(ELISA)测定培养细胞上清中MIF表达,并通过Western blot分析MIF蛋白表达。结果结肠癌标本60份中,MIF阳性19份(31.7%),正常结肠组织标本20份中,MIF阳性1份(5.0%),差异有统计学意义(P<0.05);MIF阳性组和阴性组MVD表达,2组差异有统计学意义(P<0.05)。上皮细胞系MCF-10A和结肠癌细胞HCT116、CCL229培养上清液中MIF浓度分别为(1.1±0.9)、(15.3±3.5)和(6.7±2.1)ng/ml。Western bolt分析表明肿瘤细胞较正常细胞分泌MIF明显增加。结论MIF在结肠癌肿瘤组织中广泛表达,其表达和MVD生成有明显相关,可能与结肠癌的形成和进展相关,可能成为新的治疗靶点。with angiogenesis Objective To explore the role of macrophage migration inhibitory factor (MIF), and its relationship in human colon cancer progression. Methods The expression of MIF protein was investigated by method in 60 cases of human colon cancer tissues, and the micro vascular density (MYD) was examined by using immunohistochemical CD34 staining. In addition, the expression of MIF in MDA-MB-231, MCF-7 (colon cancer cell lines) and MCF-IOA (epithelial cell line) cells was analyzed by ELISA, and the MIF protein content was detected by Western blot. Results The positive expression rate of MIF protein was 19(31.7% ) in 60 cases of colon cancer, and 1 (5.5 % ) in normal control tissues respectively. MIF was overexpressed in colon cancer tissues compared with that of normal tissues. MVD increased significantly in MIF positive group than that of negative group ( P 〈 0. 05). Conclusion MIF protein is overexpressed, and has a close relationship with angiogenesis in colon cancer, suggesting that they may play an important role in tumor genesis and development of colon cancer, and may become a new target of treatment.
