流式细胞术检测鼻腔鼻窦肿瘤细胞周期的DNA分析

摘　　要：目的探讨流式细胞术检测DNA指数(DNA index,DI)及细胞周期在鼻腔鼻窦肿瘤组织中的意义。方法取石蜡包埋的30例鼻腔鼻窦恶性肿瘤、30例鼻腔内翻性乳头状瘤标本为实验组;以30例鼻中隔偏曲患者的正常黏膜组织作为对照组。用流式细胞术检测各组的DNA指数及细胞周期的S期细胞百分数(S-phase Fraction,SPF),研究各指数与肿瘤良恶性之间相关性。结果鼻腔内翻性乳头状瘤组DI值高于对照组,但无统计学意义,而SPF值差异有显著性;恶性组织组与对照组之间,DI与SPF的差别均有高度统计学意义;恶性肿瘤组与内翻性乳头状瘤组之间,DI值、SPF值的差别均有统计学意义。结论流式细胞术检测DI、SPF在鼻腔鼻窦良、恶性病变的诊断中有重要意义。Objective To explore the significance of flow cytometric （FCM） detection of DNA index （DI） and the cell cycle in rhinosinusal neoplasm. Methods 3 0 paraffin-embedded specimens of rhinosinusal malignant tumors and 3 0 of nasal inverted papilloma were chosen as experimental group ; 3 0 specimens of normal mueosa from nasal septum deflection patients as control. The DI value and the S-phase fraction （SPF） value of cell cycle were detected in all specimens with FCM. The correlation between the indexes and the diagnosis was investigated. Results The DI value of nasal inverted papilloma specimens was higher than that of the control group but without statistical significance, while the SPF value was statistically higher than that of the control group. The DI value and SPF value of the malignant tumor specimens were significantly higher than those of the control group and nasal inverted papilloma specimens. Conclusion Flow cytometric analysis of DI and SPF may play an important role in the differential diagnosis of rhinosinusal tumors.
