咖啡因对胎鼠及乳鼠睾丸生殖细胞数量和PCNA表达的影响

摘  要:为了深入了解咖啡因(Caffeine,Caf.)对胎儿、新生儿生殖细胞数量及其PCNA(增殖细胞核抗原)表达的影响,本实验采用低(0.3mM)、中(0.6mM)、高(1.2mM)浓度Caf.体外培养SD孕18天胎鼠、0天及4天乳鼠睾丸组织块,培养时间分别为1周、2周、3周,观察Caf.对睾丸内生殖细胞数量及其表达PCNA的影响.结果如下:(1)18天胎鼠睾丸培养组织内生殖细胞数量受Caf.影响最小,0天乳鼠次之,4天乳鼠受影响较大.(2)低浓度的Caf.对生殖细胞数量影响较少;中等浓度的Caf.在培养三周后,生殖细胞的数量才减少;而高浓度的Caf.在培养二周后,生殖细胞数量已有下降,三周更明显.(3)Caf.对生殖细胞PCNA表达阳性率无明显影响,但PCNA含量在大多数实验组都有降低.由此可知高浓度Caf.长时间培养后使生殖细胞数量减少,降低了生殖细胞PCNA的含量.Low (0. 3mM) medium (0. 6mM) and high (1. 2mM) doses of caffeine were used in vitro in testicular organ culture taken from 18 days fetal rats 0 day and 4 days neonatal rats to investigate the effects of caffeine on the number and the PCNA expression of germ cells. The results were as follows:(1)The effects of caffeine on 18 days fetal testes explant were the least; thoes on the 0 day were moderate; while those on the 4 days testes were the most; (2)Low doses of caffeine exerted little influence on germ cells. Medium doses of caffeine decreased the number of germ cells after 3 weeks of culture. High doses of caffeine, however, reduced germ cell number just after 2 weeks of culture; (3)The rate of PGNA positive cells exhibited no difference after exposure to caffeine. But the degree of PCNA expression decreased in most groups. Thus it may be deduced that high doses of caffeine reduce the number of germ cells after culturing for a long time, and decrease the degree of PCNA expression
