摘 要:目的研究外源性碱性成纤维细胞生长因子(bFGF)对吸烟大鼠脑梗死体积和脑细胞凋亡的影响。方法将40只Wister大鼠随机分为正常对照组、长期大量吸烟组(香烟10支×2次/d,90d)、长期小量吸烟组(香烟5支/d,90d)、长期大量吸烟+腹腔内注射bFGF组(香烟10支×2次/d,90d;bFGF135μg/kg·w,2次/d,15d),每组10只。每组随机取5只大鼠用线栓大脑中动脉法建立脑梗死模型,采用病理图像分析法检测各组大鼠脑梗死体积;采用免疫组化法和TUNEL法检测脑内p53表达和细胞凋亡。结果各吸烟组大鼠脑梗死体积较正常对照组明显增加,其中长期大量吸烟组脑梗死相对体积加权值为0·531±0·025,长期小量吸烟组为0·478±0·034,长期大量吸烟组+bFGF组为0·492±0·018,正常对照组为0·462±0·030,各组比较有显著差异(P<0·05);各吸烟组脑内p53表达及脑细胞凋亡较正常对照组均明显增多,其中以长期大量吸烟组p53及凋亡细胞灰度值为112·796±13·183、105·596±1·472,长期小量吸烟组为128·179±1·445、112·373±2·736;长期大量吸烟+bFGF组为143·095±1·873、135·568±1·258,正常对照组为184·227±3·438、177·134±2·038,各组比较有统计学差异(P<0·01)。结论吸烟可使脑梗死体积增加,脑内p53及细胞凋亡增加,并且与吸烟剂量呈正相关。外源性bFGF可减小脑梗死体积,并通过抑制p53表达来抑制细胞凋亡。Objective To study the effect of exogenous basic fibroblast growth factor (bFGF) on cerebral infarction volume and cell apoptosis in the smoking rats. Methods 40 rats were randomly grouped into normal control, long-term massive, long-term trifle and bFGF (abdominal cavity injection) treated long-term massive groups, 10 rats for each group. 5 rats were randomly selected from each group to establish MCAO model, detect infarction volume of each group by pathological image analysis, and p53 expression and apoptosis were detected in other rats by immunohistochemistry and TUNEL method respectively. Results The infarction volume of all the smoking groups were significantly enlarged compared with that of normal group, relative volume weighted value in long-term massive, trifle, long-term massive ± bFGF and normal control groups was 0. 531 ± 0. 025, 0. 478 ± 0. 034, 0. 492 ± 0. 018 and 0. 462 ± 0. 030, respectively, having significant difference between each group ( P 〈 0.05 ). The p53 expression and apoptosis in all the smoking groups were significantly increased com- pared with those of normal group, p53 expression and apoptosis gradation in long-term massive, trifle, long-term massive ± bFGF and normal control groups were 112. 796 ± 13. 183 and 105. 596 ± 1. 472, 128. 179 ± 1. 445 and 112. 373 ±2. 736, 143. 095 ± 1. 873 and 135. 568 ± 1. 258, 184. 227 ± 3. 438 and 177. 134 ± 2. 038, respectively; having significant difference between each group (P 〈 0.01 ). Conclusions Smoking could enlarge cerebral infarction volume, increase p53 expression and apoptosis, and it has positive correlation to smoking doses. Exogenous bFGF could reduce cerebral infarction volume, and depress apoptosis by depressing p53 expression.
