摘 要:异硫氰酸荧光素(FITC)标记的紫孢侧耳单核原生质体与未标记的糙皮侧耳单核原生质体经PEG融合后,在倒置显微镜下,用显微操纵仪直接吸取一方带有荧光另一方不带荧光的原生质体对,经培养后,根据“锁状联合”这一形态特征挑选出融合子,融合率为5.2×10^(-3).进一步鉴定结果表明,融合菌株的酯酶同工酶谱发生了明显的变化,其生物量和生长速度有不同程度的提高,子实体形态也与双亲有所不同.Fluorescein isothiocyanate (FITC), which was not toxic to protoplasts, was used for the fusion of protoplasts. After the fusion of the FITC-labelled monokaryon protoplasts from Pleurotus sapidus and the non-FITC-labelled monokaryon protoplasts from Pleurotus ostreatus, a pair of sticked protoplasts was obtained with micromanipulator under an inverted microscope. Cultured with liquid regeneration medium, the fusants were selected according to the clamp connection of dikaryocyte with a fusion rate of 5.2?0^(3). It was found that the estcrase isozyme zymogram of fusants changed greatly, while the biomass and growth rate of many fusants were superior to those of their parents, and that the fruitbody morphology of the fusants was different from their parents.
