金针茹原生质体外源DNA转化

摘  要:利用电击法将金针茹(Flammulinavelutipes)金杂19菌株的染色体DNA导入金针菇088菌株的单核菌丝原生质体中,根据转化的双核菌丝具“锁状联合”这一形态学特征筛选出了转化菌株。转化率为(7.38~12.5)×10-5,相对转化率为14.0%~18.9%。转化菌株的酯酶同工酶酶谱中出现了亲株没有的新酶带,说明遗传物质发生了变异。出菇实验表明,转化菌株的菌丝生长速度快于金针菇088菌株,现蕾期提前7天左右,菇形较好,生物学效率达91.7%。In this paper,the total chromosome DNA of Flammulina velutipes strain 19 were transferred into protoplasts of monocaryon mycelia of Flammulina velutipes strain 088 by electroporation.The transformants were obtained by the clamp connection of dicaryon mycelia.The transformation frequency was(7.38~12.5)×10-5,the relative transformation frequency was 14.0%~18.9%.Analysing the esterase isozyme,we found the new non-parental bands appeared in the pattern of transformants,and it indicated that the genetical material had variated.In the phase of forming fruit body,the growth rate of the mycelia of the transformants was faster and the forming fruit body period was earlier than Flammulina velutipes strain 088,and the shape of fruit body was also better.The biological efficiency was 91.7%.
