摘　　要：目的观察8种中药有效成分对细胞色素P450酶(cytochrome P450,CYP)1A1,2E1,3A4和3A5基因表达的影响。方法采用实时定量PCR技术检测HepG2细胞中各CYP mRNA的表达。结果黄芩苷、黄芩素和青蒿素在不同浓度下均明显地诱导CYP1A1的表达。与黄芩苷和黄芩素相比,青蒿素对CYP1A1的诱导作用较弱。七叶皂苷钠、黄芩素和青蒿素能明显诱导CYP3A4的表达。结论HepG2细胞可作为CYP诱导剂的体外筛选模型。黄芩苷、黄芩素、青蒿素和七叶皂苷钠对CYP1A1或CYP3A4的诱导作用,为中西药代谢性相互作用及毒理学研究提供了实验依据。Objective To study the effects of the active components of eight kinds of Chinese herbs on cytochrome P450 enzymes （CYP） 1A1, 2El, 3A4 and 3A5 mRNA expression. Methods The mRNA expression levels of four CYP enzymes were determined by real - time quantitative reverse - transcriptase polymerase chain reaction. Results Baicalin, baicalein and artemisinin induced CYP1A1 expression at different concentrations. Compared with baicalin and baicalein, the effect of artemisinin was weaker. The expression of CYP3A4 gene was significantly obvious after induced by sodium aescinate, baicalein and artemisinin. Conclusion HepG2 cells should be an appropriate in - vitro system for investigating potential human CYP inducing agents. CYP1A1 and CYP3A4 expression could be significantly induced by baicalin, baicalein, artemisinin and sodium aescinate, which would supply the evidence for the interaction of herbal medicine and western medicine based on cytochrome P450 and toxicology.
