摘 要:目的:观察壳聚糖对白血病细胞(K562,SHI-1)的作用。方法:以不同浓度的壳聚糖处理白血病细胞K562后,运用RT-PCR方法检测实验组K562细胞中多肽:N-乙酰氨基半乳糖转移酶-2(ppGalNAc-T2)mRNA表达差异。另用不同浓度的壳聚糖分别作用于白血病K562细胞和SHI-1细胞,MTT法检测壳聚糖对两种细胞的相对抑制率。结果:与阴性对照组比较,实验组K562细胞的ppGalNAc-T2表达降低,且随壳聚糖浓度升高,表达量进一步减少;MTT实验显示不同浓度的壳聚糖对K562细胞和SHI-1细胞均有抑制作用,并呈浓度依赖性。结论:壳聚糖对肿瘤细胞有一定的抑制作用,同时还可以减少ppGalNAc-T2在mRNA水平的表达。Objective: To observe the effect of chitosan on leukemia cells (K562, SHI-1 ). Methods: The leukemia ceils were treated by different concentration of chitosan. The expression of ppGalNAc-T2 on K562 ceils were determined by RT-PCR method. The leukemia ceils were treated by different concentration of chitosan, and the relative inhibition rate was determined by MTT assay. Results: With adding the chitosan to compare with the negative group, the expression of ppGalNAc-T2 in the experimental group K562 ceils reduced, and had the inverse ratio with the chitosan's concentration. In the MTT assay, the chitosan with various concentrations had the inhibitory action on the K562 ceils and the SHI-1 ceils. Conclusion: The chitosan suppressed the K562 ceils and the SHI-1 ceils multiplication, and its action advanced along with the concentration. Chitosan influenced the expression of ppGalNAc-T2 in leukemia cell K562 reduced its expression quantity.
