摘 要:目的检测PML-RARα融合基因疫苗在小鼠体内的转录和表达。方法用成功构建的重组质粒(PML-RARα-hIL-2)免疫小鼠,于第0周、2周、4周在双侧股四头肌各注射1次,分离注射部位肌肉,用RT-PCR检测目的基因在小鼠骨骼肌中的转录;应用免疫组织化学染色法检测重组质粒蛋白表达。结果小鼠注射部位肌肉中检测到PML-RARα及hIL-2基因的转录,并可检测到hIL-2蛋白的表达。结论所构建的PML-RARα系列重组质粒可以在小鼠体内有效地表达基因产物,具有DNA疫苗的基本特性。To investigate the transcription and expression of PML - RARer fusion DNA vaccine in vivo in mice. Methods The reconstructed plasmids which were previously developed in our laboratory were injected into BALB/C mice at 14-day intervals for three cycles. The transcription and expression of PML- RARα and hIL-2 in injection site were detected by RT-PCR and immunohistological methods. Results The PML-RARα/hIL- 2 mRNA and protein were identified in the injection site of PML-RARα-hIL-2 vaccinated mice. Conclusion The reconstructed PML-RARα plasmids can effectively express gene production in the skeletal muscles of mice and have the common features of DNA vaccine.
