摘  要:目的分析PML-RARα融合基因疫苗诱导小鼠特异体液和细胞免疫应答情况。方法用成功构建的重组质粒(pIRES-PML-RARα-hIL-2)免疫小鼠后,分别利用ELISA法检测小鼠血清中INF-γ生成情况;LDH释放法检测小鼠脾细胞特异性CTL细胞针对APL细胞株NB4的杀伤率;用间接免疫荧光法检测血清中抗PML-RARα抗体。结果免疫后第7周时,小鼠血清中INF-γ含量、抗PML-RARα抗体及脾脏CTL细胞针对NB4的杀伤率均高于对照组。结论所构建的pIRES-PML-RARα-hIL-2重组质粒可诱导小鼠产生了特异性体液和细胞免疫应答。Objective To evaluate the in vivo immune effect induced by PML - RARα vaccines in mice. Methods After immunization with reconstructed pIRES-PML-RARα-hIL-2, the specific cytotoxicity of spleen cells from thevaccinated BALB/C mice was detected by LDH release assay; interferon-γ secretion was measured by ELISA; antibodies against human PML-RARα were detected by indirect immunofluorescene analysis. Results Time dependent antibody production and an increase in INF-γ were be detected in vaccinated mice serum. Specific cytotoxicities on NB4 cells were significantly higher than those in control groups. Conclusion and cellular immune The recombinant plRES-PML-RARα hIL-2 plasmids can induce specific humoral responses in BALB/c micie in vivo.
