寨卡有效部位提取物对人结肠癌LoVo细胞体外抑制作用的研究

摘　　要：目的研究寨卡有效部位提取物体外抑制人结肠癌LoVo细胞的量-效和时-效关系,并对其可能的作用机制进行初步探讨。方法寨卡有效部位提取物不同浓度、不同时间分别处理LoVo细胞,以MTT法和荧光显微镜观察其对细胞生长增殖的抑制作用,利用分光光度法检测试剂盒检测Caspase-3和Caspase-9的活化程度。结果寨卡有效部位提取物对体外培养的人结肠癌LoVo细胞有明显的抑制作用,呈剂量和时间依赖关系,IC50值为550μg/ml。Caspase-3和Caspase-9的活性与药物浓度呈正相关,当浓度为1000μg/ml时其活性与阳性药物组基本相当。结论寨卡有效部位提取物在体外能有效抑制人结肠癌LoVo细胞的生长,并可能通过激活Caspase-3和Caspase-9诱导细胞凋亡。Objective To investigate the concentration - effect and time - response effect of active fractions of Pennycress （ZAF） against human colon carcinoma LoVo cell and the possible mechanism in vitro. Methods In vitro cultured human colon carcinoma cell LoVo cells were exposed to ZAF at different concentration or different treating time, then the growth inhibiting effects of the cell were detected with MTY assay and double fluorescence staining methods, activities of Caspase -3 and Caspase -9 were studied by Assay Kit. Results Human colon carcinoma LoVo cells were inhibited more significantly with increase of ZAF concentration and prolongation of culture time, IC50 550 μg/ml. ZAF could activate Caspase - 3 and Caspase - 9 in a concentration dependent manner. When treated with 1 000 μg/ml ZAF, activation of Caspase - 3 and Caspase - 9 corresponded with anti - tumor medicine Cisplatin. Conclusion ZAF can inhibit the growth of the human colon carcinoma LoVo cells in vitro effectively, and trigger the activation of Caspase - 3 and Caspase - 9 to induce cell apoptosis.
