摘  要:目的研究实验性蛛网膜下腔出血后柔脑膜的纤维化及其观测指标。方法采用SD大鼠枕大池两次注血法建立模型,分别在3d、6d、10d、14d、21d取脑脊液检测转化生长因子-β1(TGF-β1)和Ⅰ型前胶原羧基端前肽(PICP);21d的脑组织予以冰冻切片后,行MASSON染色观察柔脑膜的胶原纤维。结果(1)实验组脑脊液中的TGF-β1较对照组明显升高并呈双时相(P<0.01)。21d柔脑膜胶原纤维较对照组增多(P<0.01);(2)实验组脑脊液中的PICP较对照组明显升高(P<0.01);(3)21d PICP与柔脑膜的胶原纤维衡量指标显著正相关(r=0.605,P<0.05)。结论SAH发生后,可引起TGF-β1的升高,柔脑膜纤维化;脑脊液中PICP可作为检测柔脑膜的纤维化程度的指标。Objective To research leptomeningeal fibrosis and its measured index after subarachnoid hemorrhage (SAH) on experimental models. Methods Rats were used to construct animal models of SAH by twice injecting autoblood solution into the cisterna magna. On the 3rd,6th, 10th, 14th and 21st day,the levels of TGF-1 and PICP in collected CSF were measured respectively. Masson staining was performed to the frozen sections of the 21st day' s brains. Using multimedia chromatic colour analytical system, we did quantitative analysis towards the leptomeningeal collagen fiber. Results ( 1 ) The levels of TGF-β1 in CSF of experimental group rose statistically significantly by double phase, compared with control group( P 〈0.01 ). On the 21th day ,leptomeningeal fibrosis of experimental group increased significantly( P 〈 0.01 ). (2) The levels of PICP in CSF of experimental group rose statistically significantly, compared with control levels (P 〈 0.01 ). (3) On the 21st day,there was a positive correlation between PICP and index of leptomeningeal collagen fiber( r = 0.605 ,P 〈 0.05). Conclusion The level of TGF-β1 would rise and leptomeningeal fibrosis would be produced in the animal models after SAH. PICP in CSF can be presumed as the index of leptomeningeal fibrosis.
