摘  要:目的观察骨髓间充质干细胞移植于大鼠缺血再灌注损伤眼的视网膜下后生长、分化和迁移的情况。方法体外培养大鼠骨髓间充质干细胞并传至第四代,建立大鼠眼缺血-再灌注损伤模型。将Brdu标记的BMSCs移植于大鼠缺血再灌注眼内的视网膜下,应用荧光免疫组织化学方法示踪移植后10、20、30天视网膜中Brdu标记的BMSCs的分布情况。结果移植后10d荧光显微镜下见大部分Brdu标记的BMSCs在移植区城周围成簇聚集。移植后20d观察发现BMSCs分布较多的位置为外核层。移植后30d可见BMSCs部分出现在节细胞层并且连接成片。在移植后整合于视网膜的BMSCs中,部分细胞显示NSE、GFAP、NF200阳性。结论移植于缺血再灌注损伤眼视网膜下的BM-SCs具有向损伤部位迁移和分化为神经样细胞的能力。Objective To explore the development differentiation and migration of the BMSCs (bone marrow mesenehymal stem cells), after the BMSCs were transplanted into subretinal space in transient isehemia-reperfusion injuries rat eye model. Methods After the BMSCs were separated and purified,they were generated to the fourth generation.Transient isehemia-reperfusion injuries rat eye model was established. Brdu labeled BMSCs were transplanted into subretinal space in transient isehemia-reperfusion injuries rat eye model for 10 days,20 days or 30 days. Immunoeytofluoreseent staining was used to detect migration and differentiation of Brdu labeled BMSCs, when the transplation was. Results 10 days after the transplantation, the Brdu labeled BMSCs were mainly crowed around the transplanted site.20 days after the transplantation, most of the Brdu labeled BMSCs were detect in the outer granular layer of retina.30 days after the transplantation, the Brdu labeled BMSCs were alignen in the cell layer. Then the positive area enlarged and scattered into bipolar layer layer and ganglion cell layer. At last more positive cell could be found migrate to the lesion site. The sequenced sections used for immunoeytofluoreseent identification expressed NSE,GFAP,NF200. Conclusion BMSCs which were transplanted into subretinalspaee in transient isehemia-reperfusion injuries rat eye model migrated to injury space and differentiated into retina-like structures.
