摘  要:目的观察白介素-1β(IL-1β)诱导小型猪冠状动脉内膜增殖时Rho激酶和MCP-1 mRNA表达与通心络干预作用。方法24头小型猪随机分为假手术组(n=8)、模型组(n=8)和通心络组(n=8)。假手术组和模型组喂养普通饲料,通心络组喂养普通饲料+通心络[1g/(kg·d)]。2周后麻醉、左侧开胸手术,选择左前降支及回旋支近端管腔外径近似相同的两处节段进行仔细分离,假手术组在血管外膜包裹吸附生理盐水纸巾,模型组和通心络组包裹吸附白细胞介素-1β(2.5μg)纸巾,术后继续以上喂养方法。2周后采用冠状动脉造影观察各组管腔狭窄情况。造影结束后处死动物,截取包裹血管段,进行病理学检查,并采用RT-PCR方法,测定各组手术处理血管段Rho激酶、MCP-1 mRNA的表达。结果冠状动脉造影显示,模型组冠脉管腔20%~30%狭窄,病理学检查可见模型组内膜增殖、炎性细胞浸润,平滑肌细胞内膜下迁移。通心络组管腔狭窄程度、内膜增埴及炎性细胞浸润现象较模型组明显减轻。通心络组Rho激酶、MCP-1 mRNA表达强度低于模型组(P<0.05)。结论IL-1β诱导冠状动脉内膜增殖时Rho激酶和MCP-1表达明显增强,通心络抑制其表达可能是其抑制冠状动脉内膜增殖和炎症反应的重要机制之一。[Objective] To observe the expressions of Rho-kinase and MCP-1 during the intima proliferation of coronary artery of mini-swine induced by IL-1β and the intervention of Tongxinluo. [Methods] 24 mini-swine were randomly divided into 3 groups: control group (n =8), model group (n =8), and Tongxinluo group (n =8). The control group and model group were fed with common diet; the Tongxinluo group were fed with common diet and Tongxinluo [1 g/(kg-d)]. Mter 2 week's feeding, chest operation was done for isolation of left-anterior descending branch and circumflex branch proximate. Coronary artery membrane externa in swine was packed by paper towel absorbing sodium chloride in control group, but by paper towel absorbing IL-1β (2.5μg) in model group and Tongxinluo group. After 2 weeks, lumens narrow was observed by coronary arteriography. The vascular section in the treated site was taken out to be examined by pathology and the expressions of Rho-kinase and MCP-1 were measured by RT- PCR. [Results] Compared with control group, luminal stricture, intima proliferation and inflammatory cells infiltration in Tongxinluo group were obviously decreased and the expression levels of Rho-kinase and MCP-1 were also lower than those of model group (P 〈0.05). [Conclusion] The expressions of Rho-kinase and MCP-1 was increased during intima proliferation of coronary artery, and Tongxinluo has obviously inhibitive effect on these expressions. Maybe that's one of the most important mechanisms to inhibit intima proliferation of coronary artery and inflammatory reaction.
