摘 要:目的:研究低氧去血清培养条件下半乳糖凝集素-1(Gal-1)对星形胶质细胞增殖和细胞周期的影响。方法:低氧去血清条件下体外培养大鼠星形胶质细胞,分别用高(10mg/L)、中(1mg/L)、低(0.1mg/L)剂量Gal-1干预。采用BrdU掺入法检测星形胶质细胞增殖,流式细胞术检测星形胶质细胞的细胞周期变化。结果:干预1d后,Gal-1高、中、低剂量组BrdU阳性细胞率均低于无干预对照组(P<0.05);干预3d后,Gal-1高剂量组BrdU阳性细胞率仍低于无干预对照组(P<0.05),中、低剂量组BrdU阳性细胞率与对照组比较,差异无统计学意义(P<0.05)。干预1d后,Gal-1高、中剂量组星形胶质细胞S期细胞比例降低,G0/G1细胞比例增高(P<0.05)。结论:Gal-1抑制星形胶质细胞的增殖和细胞周期进程,可能参与抑制脑缺血后的胶质疤痕形成。Aim :To investigate the effects of galectin-1 ( Gal-1 ) on the proliferation and cell cycle of astrocyte in serum-free and low-oxygen culture. Methods:Rat astrocyte was incubated by serum-free culture media combined with 10,1 , 0. 1 mg/L Gal-1 ,respectively. Then,astrocytes were incubated with BrdU for 1 day to detect the proliferation, and the cell cycle was analyzed by flow cytometry. Results: Compared with the astrocytes cultured without Gal-1, the proporation of Br- dU( + ) astrocytes cultured with 10,1,0.1 mg/L Gal-1 for 1 day were lower(P 〈0.05), that of astrocytes cultured with 10 mg/L Gal-1 for 3 days was still lower(P 〈0.05). Cell cycle of astrocyte cultured with 10 and 1 mg/L Gal-I for 1 day was arrested with the decrease of S-phase cells. Conclusion:Galectin-1 can reduce the proliferation of astrocyte and arrest its cell cycle progress, which may be useful to reduce the formation of astroglia scar appearing after brain ischemia.
