酸性肽对硝普钠诱导体外培养SD大鼠海马神经细胞凋亡的保护作用

摘  要:目的:观察不同剂量酸性肽(AP)对NO供体硝普钠(SNP)诱导的大鼠海马神经细胞凋亡的影响。方法:分离培养新生SD大鼠海马神经细胞,用终浓度为50μmol/L的SNP诱导海马神经细胞凋亡。实验共设空白对照组、SNP处理组和AP实验组,AP实验组分别加入浓度为0.0375、0.0750、0.1500g/L的AP与SNP共培养24h后,通过细胞形态学观察、MTT测定、吖啶橙荧光染色及DNA琼脂糖凝胶电泳等方法检测细胞凋亡情况及细胞存活率。结果:SNP处理组海马神经细胞存活率降低至58.9%±4.5%,与空白对照组比较,差异有统计学意义(P<0.01)。3个AP实验组海马神经细胞存活率均高于SNP处理组(P<0.05或0.01)。SNP组凋亡细胞的形态学改变及细胞核固缩、核碎裂现象明显,DNA琼脂糖凝胶电泳图谱具有DNAladder;各AP实验组凋亡细胞形态学改变不明显,DNA琼脂糖凝胶电泳图谱没有DNAladder出现。结论:AP对SNP诱导的海马神经细胞凋亡具有保护作用。Aim : To study the effects of acidic peptide(AP) on apoptosis of cultured hippocampal neurons induced by sodium nitroprusside (SNP). Methods: The new born SD rat hippocampal neurons were cultured in serum free medium with 50 μmol/L SNP for establishing neuronal apoptosis model. The hippocampal neurons were allocated into control group, SNP group, and three AP experimental groups. Different concentrations of AP(0.037 5, 0.075 0, and 0. 150 0 g/L) was added to AP experimental group for 24 h. Neuronal viability and morphological change were detected by MTT assay and in- verted microscope. Fluorescent staining and DNA agarose gel electrophoresis were used to analyze apoptosis. Results: The neuron survival rate in SNP group decreased to 58.9% ±4.5% , which was lower than that in control group (P 〈 0.01 ), The neuron survival rates in three AP experimental groups were higher than that in SNP group(P 〈0.05 or 0.01 ). AP improved thc morphologic changes and prevented the neuronal neclei from pyknosis, condensation, and fragmentation triggered by SNP. It also blocked neuroual nuclear DNA fragmentation elicited by SNP. Gonclusion :AP could protect neurons from apoptosis iuduced by SNP.
