摘 要:目的:检测人大肠癌组织中耐药基因ABCG2mRNA的表达并探讨其与临床病理特征的关系。方法:采用RT-PCR方法检测102例大肠癌组织(男60例,女42例;≥60岁63例,<60岁39例;乳头状腺癌21例,管状腺癌48例,黏液腺癌13例,腺鳞癌20例;肿瘤直径≥5cm者58例,<5cm者44例;高分化23例,中分化49例,低分化30例;有淋巴结转移72例,无淋巴转移30例;Duke's分期:A期14例,B期20例,C期42例,D期26例)及15例正常大肠黏膜组织中ABCG2mRNA的表达。结果:大肠癌和正常黏膜组织中ABCG2mRNA的阳性表达率分别为58.8%和0(P<0.05)。大肠癌组织中ABCG2mRNA的阳性表达率与患者的性别、年龄、肿瘤大小、组织学类型、淋巴结转移情况、Duke's分期等均无关(P均>0.05),与分化程度有关(P<0.05)。结论:ABCG2的表达可以作为判断大肠癌分化程度及预测化疗效果的指标之一。Aim :To detect the expression of ATP-binding cassette superfamily G member 2 ( ABCG2 ) mRNA in human colorectal carcinoma tissue, and explore its relationship with clinicopathological feature. Methods: RT-PCR method was used to detect the ABCG2 mRNA expression in 102 cases of human colorectal carcinoma (60 cases of male, 42 cases of female ;63 cases of t〉60 years old, 39 cases of 〈 60 years old; 21 eases of papillary adenocarcinoma, 48 cases of tubular ad- enocarcinoma, 13 case of mucous adenocarcinoma,20 cases of adenocarcinoma; tumor size : 58 cases of t〉5 cm, 44 cases of 〈 5 cm; 23 cases of well differentiated, 49 cases of moderate differentiated, 30 cases of poorly differentiated; 72 cases with lymph node metastasis, 30 cases without lymph node metastasis; Duke~ stage: 14 cases of stage A, 20 cases of stage B, 42 cases of stage C, 26 cases of stage D)and 15 cases of normal intestinal mucosa tissue. Results :The positive expres- sion rate of ABCG2 mRNA in human colorectal carcinoma and normal intestinal mucosa was 58.8% and 0 ( P 〈 O. 05 ). The positive expression rate of ABCG2 mRNA in human colorectal carcinoma tissue was not correlated with age, gender , tumor size, bistologic types, lymph node metastasis or Duke's stage ( P 〉 O. 05 ), and was correlated with differentiation grade ( P 〈 O. 05). Conclusion: ABCG2 may be an marker to judge differentiation of human colorectal carcinoma and to predict the result of chemotherapy.
