摘  要:目的:筛选具有生物转化活性的微生物,探讨其对左旋四氢巴马汀和四氢小檗碱的生物转化。方法:在培养体系中加入底物后继续培养60h,发酵液经氯仿萃取后采用硅胶柱层析分离纯化,根据MS、1H-NMR数据鉴定产物的结构。结果:分离得到轮枝霉菌(Verticilliumsp.WJ-03)对左旋四氢巴马汀的转化产物为巴马汀,对四氢小檗碱的转化产物为小檗碱。结论:首次报道了轮枝霉菌对生物碱的生物转化,发现轮枝霉菌对左旋四氢巴马汀和四氢小檗碱的生物转化反应类型为脱氢反应,而该反应与原小檗碱类生物碱的生物合成途径一致。AIM: To investigate the biotransformation of L-tetrahydropalmatine and tetrahydroberberine through screening 46 strains. METHODS: Substrates were added into the cultures of Verticillium sp. and then incubated for 60h.The culture was extracted with CHCI3 and then subjected to silica gel chromatography. RESULTS: Two metabolites were isolated and their structures were identified as palmatine and berberine on the basis of MS and 1H NMR data. CONCLUSION: This is the first report that Verticillium sp. is used for the biotransformation of alkaloids, and the selective dehydrogenation on the C tings of L-tetrahydropalmatine and tetrahydroberberine is similar to that in the biosynthetic pathway of protoberberine alkaloids.
