摘 要:目的观察胚胎神经上皮干细胞在体外克隆增殖和诱导分化的情况。方法取孕11 d大鼠的胚胎,经分离获得神经管上皮细胞,在无血清培养基内克隆、增殖、传代。取不同时期的神经球进行神经干细胞的鉴定和诱导分化,并通过免疫组织化学染色鉴定分化的结果。结果胚胎神经上皮干细胞在体外能够克隆、增殖,形成的神经球呈nestin阳性,在分化培养基内可分化为星型胶质细胞和神经元。结论胚胎神经上皮干细胞可在体外进行单细胞克隆和诱导分化。Objective To observe the amplification and differentiation of the embryonic neuroepithelial stem cells in vitro. Methods The neuroepithelial stem cells were obtained from the E11d embryos,and cultured in the free-serum medium: These cells were identified and induced during different culturing periods in vitro by immunocytochemistry. Results The embryonic neuroepithelial stem cells could amplify through cloning and the neurospheres were nestin-positive. The neurons and astrocytes were observed in the differentiation medium. Conclusion The embryonic neuroepithelial stem cells can grow and amplify by single cloning and differention by inducing in vitro.
