摘  要:目的探讨在缺血后给予赤芍801对脑缺血/再灌注损伤的保护作用及可能的抗氧化作用。方法大鼠局灶性脑缺血/再灌注损伤后,通过NSS和运动功能评分观察赤芍801对大鼠神经功能的影响。分别通过TTC染色、Nissl染色和TUNEL染色观察再灌注后脑梗死体积、缺血周边区神经元的存活和凋亡情况。检测缺血周边区MDA和SOD指标,探讨赤芍801发挥神经保护的可能机制。结果缺氧后给予94μmol·kg-1·d-1赤芍801能很好的改善大鼠的NSS和运动功能评分,减少缺血周边区TUNEL阳性细胞数,增加Nissl染色阳性细胞数,并减少MDA的生成、提高SOD的含量。结论赤芍801能通过抗氧化作用,保护脑缺血周边区的神经元,从而提高脑缺血/再灌注大鼠远期的神经功能恢复。Aim To investigate the protective effect of the proply gallate against cerebral ischemia-reperfusion injury and the mechanism linked to antioxidation. Methods The focal cerebral ischemia-reperfusion injury was performed by reperfusion after 2 h occlusion in rat right middle cerebral artery, and the proply gallate (94 μmol·kg^-1·d^-1) was injected immediately after carotid occlusion. The neurological severity was evaluated by the neurological severity scores(NSS) and the score of motion function. The infarct volume, the viability or apoptosis of neurons in the peri-ischemic region were detected by the TTC, Nissl and TUNEL staining respectively. The antioxidation was assessed by the level of MDA and SOD. Results The proply gallate significantly improved NSS and the score of motion function in rats, reduced the positive cells of TUNEL staining, and increased the positive cells of Nissl staining, with less production of MDA and higher activity of SOD. Conclusion The proply gallate could protect the neuron in the peri-ischemic region from cerebral ischemia-reperfusion injury by increasing the antioxidation,thus improved the long-term neurological outcome in rats.
