摘  要:目的:探讨缺氧诱导因子-1α(HIF-1α)和凋亡抑制蛋白survivin在胃癌发展过程中的意义及其对肿瘤血管生成的影响.方法:应用组织芯片技术和免疫组织化学方法检测HIF-1α和survivin在70例胃癌组织、14例癌旁组织和15例正常组织中的表达,并计数CD105标记癌组织的微血管密度(MVD).结果:HIF-1α和survivin在胃癌组织中的表达明显高于正常组织及癌旁组织(P<0.05);HIF-1α和survivin的表达均与胃癌的浸润深度、临床分期及局部淋巴结转移之间存在相关性(P<0.05);HIF-1α和survivin表达阳性组的MVD均高于各自的阴性组(P<0.05);HIF-1α和survivin在胃癌中表达存在正相关性(r=0.285,P<0.05).结论:HIF-1α和survivin在胃癌的发展和转移过程中具有重要意义;联合检测两者的表达可用于评价胃癌的恶性生物学行为.AIM: To investigate the roles of hypoxia inducible factor-1 alpha (HIF-1α) and survivin in development of gastric cancer and their effects on tumor angiogenesis. METHODS: The expression levels of HIF-1α and survivin proteins in pathological specimens from 70 cases of gastric cancer, 14 cases of pericancerous tissue and 15 cases of normal tissue were examined by tissue microarray technique and immunohistochemistry. The CD105 labeled micro vessel density (MVD) was counted in each group. RESULTS: The expression levels of HIF-1α and survivin were significantly higher in gastric cancer tissue than in pericancerous tissue and non-tumor tissue ( P 〈 0.05 ). The over expressions of HIF-1α and survivin were strongly related to the invasiveness, clinical stages and metastasis of gastric cancer ( P 〈 0.05 ). The MVD in HIF-1α or survivin positive group was higher than that in HIF-1α or survivin negative group. The expression of HIF-1α was positively correlated with the expression of survivin ( r = 0. 285, P 〈0.01 ). CONCLUSION: HIF-1α and survivin proteins play critical roles in carcinoma metastasis and biological behavior of gastric cancer. Co-detection of HIF-α and survivin expression can be used to evaluate some biological behaviors of gastric cancer.
