摘  要:目的探讨肝内胆管癌组织中缺氧诱导因子-1(HIF-1α)的表达及其与VEGF的表达、微血管密度(MVD)及临床病理因素之间的关系。方法采用免疫组化方法检测56例胆管癌及20例癌旁正常肝内胆管中HIF-1α、VEGF的表达,用CD34单克隆抗体标记血管内皮细胞并计数MVD,分析其间的相关性及其与肿瘤临床病理特征之间的相关性。结果肝内胆管癌组织中HIF-1α和VEGF的阳性表达率显著高于正常组织(P<0.05);HIF-1α和VEGF的表达均与淋巴结转移和TNM分期呈正相关(P<0.05);HIF-1α的表达与VEGF的表达和MVD值呈正相关(P<0.05)。结论HIF-1α在胆管癌组织中明显上调,与肿瘤血管生成、肿瘤转移及预后密切相关;提示检测HIF-1α可能成为判断胆管癌生物学行为及预后的重要指标,并作为临床控制胆管癌的作用靶点。[Objeelive] To investigate the expression of HIF-1α gene and its relation to vascular endothelial growth faetor (VEGF) protein and mierovessei density (MVD) in human eholangioeareinomas (ChC). [Methods] The expression of HIF-1α and VEGF were asessed by immunohistoehemieal staining in 56 eases with human eholangio- eareinomas (ChC) and in 20 eases with surrounded normal tissues, MVD was determined by anti-CD34 Endoglin im- munohitoehemieal staining and then we studied the relationship between the expression and the elinie opathologieal feature. [Results] The expressions of HIF-1α and VEGF in ChC were significantly higher than those in normal tissues (P 〈0.05). The expression of HIF-1α and VEGF were both eorrelated with lymph node involvement (P 〈0.05) and elinleal stage (P 〈0.05); the expression of HIF-1α was signifieanfly eorrelated with the VEGF expression and MVD (P 〈0.05). [Conclusions] The upregnlation of the expression of HIF-1α in ChC had a close relationship to tumor angionesis, earnogenesis, development and infdtration. The overexpression of HIF-1α and VEGF can be used as important indiees for evaluatinge biologieal behaviors and prognosis of HIF-1α, and may beeome a new target for tumor treatment.
