摘 要:目的探讨黄芪赤芍对大鼠肝纤维化纤维形成的抑制作用及机制。方法以40%四氯化碳和5%乙醇制备大鼠肝纤维化动物模型,随机分为5组后,黄芪组、赤芍组、黄芪赤芍组分别给予黄芪提取物、赤芍提取物和黄芪赤芍合剂干预后,测定血清中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、透明质酸酶(HA)、层黏蛋白(LN)水平;测定肝组织中超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量;观察肝组织中LN的表达情况。结果黄芪赤芍组与其他组相比较:血清AST、ALT、HA、LN水平明显降低(P<0.01)肝组织中SOD活性显著升高(P<0.01),MDA含量显著降低(P<0.01);肝组织中的LN表达明显减少,纤维化程度明显改善。结论黄芪赤芍合剂可减缓肝纤维化的发生,其作用机制可能与其抗氧化作用及减少LN的产生有关。[Objective] To investigate the inhibitive effect and possible mechanism of Astragalus and Paeoniae rubra misture on CCl4-ethanol-indueed hepatic fibrosis rats. [Methods] Rats were randomly allocated into five groups (nomal control group, model control group, Astragalus group, Paeoniae rubra group and Astragalus and Paeo- niae rubra misture group).CCl4 and ethanol were used to induce hepatic fibrosis. Astragalus, Paeaniae rubra and As- tragalus and paeoniae rubra misture (0.2 mL/10 g) was administered to the last three groups for 10 weeks. Model control group received saline. All groups were compared the lever of ALT, AST, A/G, HA, LN, SOD and MDA in terms of serum and tissue. [Resldtes] Compared with other groups, treatment of rats with Astragalus and Paeoniae rubra misture remarkably decreased serum AST, ALT, HA and LN levels(P 〈0.01) and liver MDA level(P 〈0.01). But increased levels of liver SOD(P 〈0.01). Meanwhile, Microscopic studies revealed that the livers of rats receiving Astragalus and Paeoniae rubra misture showed decreases in fibrosis and in expression of LN. [ Conclusions] Astragalus and Paeoniae rubra misture has inhibitive effect on experimental hepatic fibrosis. The mechanisms possibly associated with the action of antioxidant, the expression of decreasing LN.
