摘  要:目的:探讨酪氨酸激酶Syk在肝细胞性肝癌组织中的表达及与肿瘤微血管密度(microvessel density,MVD)之间的关系。方法:采用RT-PCR法检测Syk mRNA在32例肝细胞性肝癌组织及癌旁正常组织中的表达,免疫组化SABC法检测标本中CD34的表达,通过CD34的表达反映肿瘤的MVD。结果:在24例癌旁正常组织中,Syk mRNA均表达阳性,32例肝细胞性肝癌组织中表达率为46.9%(15/32),低分化组阳性表达率23.1%(3/13)明显低于高分化组阳性表达率63.2%(12/19),P=0.0329。低分化组MVD(Ⅲ和Ⅳ级分别为49.2±3.6和54.9±4.3)高于高分化组(Ⅰ和Ⅱ级分别为13.6±4.5和32.3±3.2),与正常组织(5.9±1.7)差异有统计学意义,P=0.00645;Syk mRNA的表达与CD34的表达明显负相关,r=-0.87,P=0.0026。结论:Syk基因的缺失在肝细胞性肝癌的恶性进展及血管生成中起了重要的作用。OBJECTIVE: To study the expression of Syk gene in hepatocellular carcinoma and the relationship between Syk mRNA and microvessel density (MVD) in the tumors. METHODS: The expression of Syk gene was examined with RT-PCR in 24 specimens of normal tissues surrounding the tumors and 32 specimens of hepatocellular carcinoma, and tumor's MVDs through the expression of CD34 were examined with the immunohistochemical SABC method in the same specimens. RESULTS: Twenty-four specimens of normal tissues surrounding the tumors were positively expressed of Syk gene, only 15 specimens in 32 specimens of hepatocellular carcinomas were positively expressed of Syk gene, and the lower differential HCC showed a lower positive rate (23.1%, 3/13), the high differential HCC showed a high positive rate (63. 2%,12/19), P =0.032 9, the detection of the tumor's MVD through CD34 in lower differential HCC(Ⅲ ,49.2±3.6; Ⅳ,54.9 ± 4.3) showed higher than the high differential HCC ( Ⅲ, 13.6±4.5; Ⅳ 32. 3 ± 3. 2), and compared with the nomal tissues (5.9±1.7) possessed a statistical significance, P =0. 006 45. The expression of Syk gene was negatively correlaed with CD34 completely, r=0.87, P=0. 002 6. CONCLUSION: Syk gene deletion might be responsible for tumor malignant progression and angiogenesis in hepatocellular carcinomas.
