摘  要:目的:探讨三磷酸肌醇(IP3)和bax基因表达变化在genistein治疗裸鼠移植人肝癌中的作用。方法:以裸鼠移植人肝癌为对照,对照组腹腔注入含0.04% DMSO的RPMI 1640培养基0.05mL/g,Genistein治疗组腹腔注入genistein 1mg/(kg·d),3周后观察肝癌增长情况,并应用同位素试剂盒检测肝癌组织IP3含量,RT-PCR分析癌组织bax mRNA表达,Western blotting分析肝癌组织bax蛋白表达。结果:Genistein治疗组肝癌体积和重量均显著低于对照组[体积(12.6±11.6)mm3vs(52.3±26.5)mm3,重量(42.7±27.8)mgvs(91.3±31.4)mg),P<0.01],IP3含量显著低于对照组[(13.4±1.4)pmol/mg proteinvs(35.3±6.6)pmol/mg protein,P<0.01],bax mRNA表达显著高于对照组[灰度与面积之积的相对强度(RI)0.88±0.21vs0.56±0.15,P<0.05],bax蛋白表达显著高于对照组[RI2.86±0.80vs1.37±0.48,P<0.05]。结论:Genistein能减少IP3生成,上调肝癌组织bax基因表达,抑制裸鼠移植人肝癌增长。Objective To probe into the effects of genistein on 1,4,5-trisphosphate inositol(IP3) and bax gene expression in mude mice with hepatocellular carcinoma. Methods Eighteen nude mice with hepatocellular carcinoma were randomly devided into control group (Group C, n=9) and genistein treatment group (Group G, n=9). Group G was treated with genistein 1 mg/(kg·d) (ip) and Group C with 0.04% DMSO RPMI 1640 medium 0.05 mL/g for 3 weeks. The volume and weight of tumor was then measured, IP3, bax mRNA and bax protein of hepatomatic tissues were assayed by IP3 - [ ^3H ] Birtrak assay, RT-PCR and Western blotting, respectively. Results The tumor volume and weight of animals was lower in Group G than that in Group C [ ( 12.6 ± 11.6) mm^3 vs (52.3 ± 26.5) mm^3, (42.7 ± 27.8) mg vs (91.3 ± 31.4) mg), P 〈 0.01 ] ,IP3 content was lower in Group G than that in Group C[( 13.4 ± 1.4) pmol/mg pro vs (35.3 ± 6.6) pmol/mg pro,P 〈 0.01 ], bax mRNA expression was higher in Group G than that in Group C [ R1 which was the gray degree multiply area of bax / the gray degree multiply area of β-actin 0.88 ± 0.2 vs 0.56 ± 0.15, P 〈 0.05], bax mRNA expression was higher in Group G than that in Group C(2.86 ± 0.80 vs 1.37 ± 0.48, P 〈 0.05). Conclusion Genistein can inhibit the growth of hepatocellular carcinoma tansplanted into liver of nude mice by reducing IP3 production and up-regulating bax gene expression.
