摘 要:目的探讨EphB2受体在胃癌组织、癌前病变中的表达及对血管生成和肿瘤生物学行为的影响。方法应用链霉素亲生物素-过氧化酶免疫组织化学法(S-P)检测75例胃癌组织、45例癌前病变中EphB2的表达情况并与30例正常胃黏膜组织对照,采用CD34标记血管内皮细胞,计算微血管密度(MVD)。结果EphB2在胃癌组织及癌前病变表达高于正常胃黏膜组织,分别为64.0%、33.3%和10.0%,三者差异存在显著性(H=29.113,P<0.001)。EphB2在胃癌组织不同临床病理分期表达不同,分别为0、33.3%、52.2%、80.0%和100.0%,随临床病理分期增加表达率呈上升趋势(H=23.518,P<0.001)。CD34(以MVD表示)在胃癌组织、癌前病变及正常胃黏膜表达分别为67.61±13.13、33.92±3.60和21.29±3.28,呈下降趋势(F=318.831,P<0.001)。EphB2与CD34(以MVD表示)在胃癌组织中皆呈高表达,两者呈正相关(rs=0.915,P<0.001)。结论EphB2在胃癌组织中表达并随临床病理分期增加表达率呈上升趋势,EphB2在部分癌前病变中表达,EphB2参与肿瘤血管形成。Objective To investigate expression of EphB2 receptor in human gastric cancer and precancerous lesion and its effect on angiogenesis and metastasis. Methods Expression of EphB2 was determined by immunohistochemical method( S-P method) in 75 cases of gastric cancer specimens and 45 cases of precancerous lesion specimens. Thirty normal gastric tissue specimens were selected as control group. CD34 -labelled endothelial cells were used to calculate microvessel density (MVD). Results The positive expression rates of EphB2 in gastric cancer and precancerous lesion were higher than that in normal gastric tissue, and were 64. 00%, 33.33% and 10. 00% respectively, with significant statistical difference (H = 29. 113, P〈0. 001). The positive expression rates of EphB2 in gastric cancer of different stages were 0.00%, 33. 33% ,52. 17% ,80. 00% and 100. 00%, with a tendency of increase with the advancement of stages of ( H = 23. 518, P 〈 0. 001 ). Expression of CD34 (MVD) in gastric cancer, precancerous lesion and normal gastric tissue was respectively 67. 61 ± 13. 13,33. 92 ±3. 60 and 21.29 ± 3. 28, with a falling tendency (F = 318. 831 ,P 〈0.001 ). The positive expression rate of EphB2 and CD34(MVD) were upregulated in gastric cancer,showing positive correlation between expression of EphB2 and CD34( rs = 0. 915 ,P 〈 0. 001 ). Conlusion EphB2 is overexpressed in gastric cancer and has a tendency of increase with the advancement of stages. EphB2 is overexpressed in some precancerous lesion. EphB2 contributes to angiogenesis in gastric cancer.
