AIM: To investigate the effect of curcumin on the expression of vascular endothelial growth factor (VEGF) in hepatocellular carcinoma cell line HepG2 under hypoxic conditions. METHODS: Hepatocellular carcinoma HepG2 cells were treated with different concentra- tions (0, 1, 2, 5, 10μmol/L) of curcumin under hypoxic conditions for 6 h. Cellular viability was detected by MTT assay. The level of VEGF expression was detected at protein and mRNA level by Western blot technique and reverse transcription-poly chain reaction respectively. RESULTS: The levels of VEGF protein and mRNA decreased significantly in the cells co-incu- bated with curcumin at 1, 2, 5, and 10 μmol/L for 6 h in a dose-dependent manner as compared with those in the control cells (0 μmol/L curcumin treatment) (VEGF protein: 2.12 ± 0.23,1.59 ± 0.13, 0.82 ± 0.11, 0.33 ± 0.05 vs 2.85 ± 0.37, P 〈 0.05 or P 〈 0,01; VEGF mRNA: 0.60 ± 0.05, 0.54 ± 0.04, 0.16 ± 0.02, 0.06 ± 0.01 vs 0.81 ± 0.07, all P 〈 0.01). CONCLUSION: Curcumin can decrease the expression of VEGF in hepatocellular carcinoma HepG2 cells.
