摘 要:目的探讨金粉蕨素抗人卵巢癌(COC1)细胞系细胞作用及机制。方法MTT比色法测定金粉蕨素对COC1、顺铂耐药亚株COC1/DDP及中国仓鼠卵巢(CHO-K1)细胞系细胞增殖的影响;PI染色流式细胞术(FCM)分析金粉蕨素对COC1、顺铂耐药亚株COC1/DDP及中国仓鼠卵巢(CHO-K1)细胞系细胞凋亡和细胞周期的影响。结果金粉蕨素对COC1、COC1/DDP细胞生长具有抑制作用呈剂量-效应和时间-效应关系,PI染色FCM分析发现金粉蕨素明显使COC1细胞周期停滞于G1期(G1期细胞百分数由50.3%上升到70.6%,P<0.05)。结论金粉蕨素具有体外抗人卵巢癌(COC1)细胞系细胞作用,其机制可能与细胞周期G1期阻滞有关。Objective To study the effects and mechanism of the onychin on human ovarian cancer(COC1 ) cello. Methods MTT assay was used to determine the effect on proliferation of COC1 , COC1/ddp and CHO -K1 cell by onychin;PI stained flow eytometry (FCM) was used to analyse the apoptosis and cell eyele change of COC1 treated by Onyehin. Results Onyehin inhibited COC1 and COC1/DDP cell growth signifieantly in a dose - dependent manner and a time - dependent manner. Analysis by flow cytometry showed that onychin suppressed cell cycle progression at Gi phase. 9 ( the percentages of cells iu G1 phase rose from 50. 3% to 70.6% ). Conclusion Onyehin has anti - ovarian carcinoma effect in vitro; the mechanism might be associated with G1 phase cell cycle arrest.
