摘  要:目的:观察尿毒清胶囊及其拆方对5/6肾切除法制作的慢性肾功能衰竭大鼠模型肾组织TGF-β1、FN、LN表达的影响。方法:5/6肾切除法建立慢性肾功能衰竭大鼠模型分为黄芪组、丹参组、大黄组、黄芪丹参大黄组、尿毒清组、空白对照组,空白对照组予以蒸馏水灌胃,其他组给予相应的药物煎剂灌胃干预6周。治疗前后,检测各组大鼠血尿素氮、血肌酐的水平;并采用免疫组化SABC法检测各组大鼠肾组织TGF-β1、FN、LN的表达情况,并进行定量分析。结果:TGF-β1、FN、LN在各组均有表达,上述指标在空白组表达最为强烈,与其他各组比较,差别均有显著性意义(P<0.01或P<0.05)。尿毒清组表达最轻,黄芪大黄丹参组次之。尿毒清组与黄芪组、丹参组、大黄组比较,差别有显著性意义(P<0.01或P<0.05)。其中,尿毒清组TGF-β1表达量低于黄芪大黄丹参组,差别有显著性意义(P<0.01)。结论:中药治疗CRF的分子作用机理之一,可能与通过下调TGF-β1、FN、LN在肾组织中的表达,减少ECM的聚集,减轻或延缓肾脏纤维化,保护肾功能有关。而中药复方尿毒清胶囊在下调TGF-β1、FN、LN在肾组织中的表达作用要比单味中药好。Objective :To observe the effects of Niaoduqing Capsule and its ingredients on the expressions of transforming growth factor 1 (TGF - 1 ) ,fibronectin(FN) ,laminin(LN) in renal tissue of chronic renal failure (CRF) rats modeled by 5/6 nephrectomy. Methods : CRF rats modeled by 5/6 nephrectomy were divided into 6 groups. All groups were treated by intragastric adu-nistration for 6 weeks - Group A with Radix Astragali apozem, Group B with Radix Salviae Milfiorrhizae apozem, Group C with Radix et Rhizoma Rhei apozem, Group D with apozem of Radix Astragali, Radix Salviae Mihiorrbizae and Radix et Rhizoma Rhei, Group E with apozem of Niaoduqing Capsule, and Blank Control Group with distilled water. Before and after treatment,blood urea nitrogen (BUN) and serum creatinine(SCr) were examined in all groups; the expression levels of TGF- 1 ,FN, LN in renal tissue were examined in all groups by immunohistochemical SABC method, and then were analyzed quantitatively. Results : Expressions of TGF - β1, FN, LN could be tested in all groups, Their expressions in Blank Control Group were significantly higher than those of all the other groups (P 〈 O. O1 or P 〈 O. 05 ). Group E had the lowest expressions, followed by Group D. The expressions in Group E were significantly lower than those of Group A,B ,C( P 〈 O. O1 or P 〈 O. 05 ). The expression of TGF -β1 in Group E was significandy lower than that of Group D ( P 〈 O. 01 ). Conclusion : One of the molecular action mechanisms in curing CRF by traditional Chinese medicine may be relevant to the down regulation of the TGF -β1 ,FN,LN expression levels in renal tissue which thereby reduces the gather of ECM,lessens or slows down the renal fibrous degeneration, and finally protects renal function. As regard to the down regulation of the TGF-β1 ,FN,LN expression levels in renal tissue,the complex prescription of the traditional Chinese medicine Niaoduqing Capsule has better effect than its ingredients do.
