摘  要:目的:观察脑缺血后抑郁模型大鼠下丘脑酪胺酸羟化酶(TH)表达及益肾调气方药的干预作用。方法:建立卒中后抑郁大鼠模型基础上,采用免疫组化ABC法观察脑缺血后抑郁大鼠脑内TH的表达及益肾调气方药的干预作用。结果:模型大鼠下丘脑TH阳性细胞的面密度及光密度较正常降低(P<0.05;P<0.01);对照组和治疗组TH阳性细胞面密度和光密度均较模型增加(P<0.05;P<0.01)。结论:卒中后抑郁模型大鼠存在脑内单胺递质合成催化酶-TH功能的下降,与脑内单胺递质含量下降有关,益肾调气方药可通过促进TH的单胺递质合成功能达到治疗作用。Objective:To observe the expression of Tyrosine Hydroxylase(TH) of Hypothalamus in post - stroke depression rat model and effects ofYishen Tiaoqi decoction. Methods :Models of post - stroke depression rat were estabished first, and the expression of TH of Hypothalamus in post - stroke depression rat model and effects of Yishen Tiaoqi decoction on the Tyrosine Hydroxylase of Hypothalamus were observed with avidin - blothin- peroxidase complex method of immunohistoehemisy. Restdts-The area density and Optical density (OD) of TH positive ceils of Hypothalainus in model group were all decreased than those of Normal group (P 〈 0. 05;P 〈 0. 01 ) , while the area density and OD of TH positive eeUs in control group and the treatment group were all increased than those of model group( P 〈 0. 05 ;P 〈 0. 01 ). Conclu- sion : The funeion of TH has decreased in post - stroke depression, which maybe the cause of the declines of contents of the monoainine in brain. The therapeutic effect of Yishen Tiaoqi decoction on post - stoke depression maybe the enhancement of the synthesis of monoamine in brain.
