摘  要:从长期受乙酰甲胺磷污染的老菜地土壤中,通过富集培养,分离筛选出5株乙酰甲胺磷降解菌株。选取其中1株有较强降解能力的乙酰甲胺磷降解菌XP-3,提取该菌总DNA进行16S rDNA PCR扩增、测序,BLAST检索及序列分析表明,XP-3菌与金黄杆菌属的同源性为98%。并对其进行了形态和生理生化鉴定,将其鉴定为Chryseobacterium sp.XP-3。通过进一步研究XP-3菌菌株对乙酰甲胺磷的耐药能力及降解效能表明,该菌具有较强的耐药能力,可以在1 500 mg/L浓度下生长繁殖。能以乙酰甲胺磷作为唯一的碳源和氮源生长。接种该菌悬液1mL于500 mg/L、800 mg/L的乙酰甲胺磷无机盐液体培养基中,28℃、120 r/min振荡培养24 h,对乙酰甲胺磷降解率分别可达87.76%和87.16%。Five strains of acephate degrading bacteria were isolated from the soils in which acephate was used for a long period. The preliminary study of XP-3 strain selected from them was carried out. Total DNA of XP-3 was extracted, and 16S rDNA was amplified and sequenced. Blast analysis showed that the 16S rDNA of XP-3 strain was 98 % similarity with Chryseobacterium sp. The results including biochemical tests indicated that XP-3 strain was belonged to Chryseobacterium sp. XP-3. The highest endurat ionconcent rations of the bacterium to acephate through further researchs were 800,1000,1500 mg/L respectively. It has strong ability of growth and reproduction in the 1500 mg/L acephate.This strain can use acephate as the sole carbon and nitrogen sources.In the culture conditions of 28℃ and 120 r/min, whith in 24 hours, the rates of acephate biodegradation is up to 87.76 % and 87.16 % respectively on the condition of 500 mg/L , 800 mg/L acephate .
