摘 要:应用细胞转化和核酸杂交技术研究 c-myc 癌基因与宫颈癌的关系,可见宫颈癌DNA 转染 NIH/3T3细胞,在 G_(418)-DMEM 选择性培养液中形成能持久增殖、呈重叠生长的转化灶,转化灶细胞基因组中 c-myc 癌基因有扩增现象;经斑点杂交检测正常宫颈、慢性宫颈炎和宫颈癌组织DNA 中 c-mye 癌基因,揭示宫颈癌组织中其拷贝数明显增加;宫颈腺癌 DNA 经 Hind Ⅲ单酶切进行Southern Blot 杂交出现两条阳性杂交带,而宫颈鳞癌 DNA 经 EcoRⅠ或 HindⅢ/EcoRⅠ或 EeoRⅠ酶切,均只出现一条阳性杂交带。Investigate was the relation of oncogene c-myc and uterine cervix with cell transformation and nu- cleic acid hybridization, the results showed that NIH/3T3 cells could be transformed by DNA of cervi- cal carcinoma, and the transformed loci exhibited a permanent, overlapping proliferative feature in Ge- neticin(G418)-DMEM selective medium. Oncogene c-myc amplification was observed in the transfor- mant DNA genome;then we detected the oncogene c-myc in normal cervix, chronic cervicitis and cer- vical carcinoma by dot blot hybridization;its copy in cervical carcinoma highly incerased, cervical ade- nocarcinoma DNA was treated by restriction endonuclease Hind Ⅲ and studied by southern Blot hy- bridization, two positive bands of oncogene c-myc appeared, but DNA of squamous cervical carcino- ma cell was treated by the restriction of endonuclease EcoR Ⅰ or Hind Ⅲ/EcoRI,EcoR1 and studied uncler the same condition;it only one positive hand occurred.
