人宫颈癌DNA中c-Ha-ras转化基因的检测和分析

摘  要:以同位素标记的 PT_(24)Ha-ras 癌基因为探针,采用斑点杂交方法检测20例宫颈癌、12例宫颈炎、1例恶性葡萄胎 DNA 中 c-Ha-ras 基因的同源序列,杂交结果为:20例宫颈癌均为阳性。3 例宫颈炎和1例恶性葡萄胎出现阳性信号。以基因转染技术证实了宫颈癌 DNA 的转化活性,并证明转化细胞株中 c-Ha-ras 的存在。本研究结果提示,c-Ha-ras 癌基因是人富颈癌的转化基因之一。Preparation of DNA isolated from 20 untreated cervical cancer biopsies(stage Ⅱ~Ⅲ)and 13 cervitis by hysterectiomy was examined for the state of the oncogene c-Ha-ras.Blot hybridization ex- periment demonstrated the presence of homologoues of the Ha-ras in the 20 biopsies and 4 of 13 hys- terectomies.The positive hybridization ratio for the former was 100%,for the latter about 30%. DNA from the cervical cancer biopsies induced transformed foci in cultures of NIH 3T3 cells by the calcium phosphate method.The morphologically transformed cells could grow in colony on soft a- gar.These transformants were tumorigenic in nude mice.Hybridization of DNA from transformants digested with restriction endonuclease BamH I 6.6 kb with human Alu repeat sequence was positive. Thus,the presence of the activated oncogene in tumor tissue suggests that it may play an important role in cervical cancer.
