摘 要:目的观察术中静脉应用Rapamycin及供体脾细胞对移植心脏生存时间的影响。方法建立大鼠异位心脏移植模型,术中静脉应用Rapamycin及供体脾细胞。术后通过腹壁触诊判断移植心脏存活情况。结果Rapamycin组移植心脏的生存时间为(12.3±3.9)d,与未用药组(8.5±2.4)d相比差异有统计学意义(P=0.018<0.05)。Rapamycin+供体脾细胞组移植心脏生存时间是(12.4±3.7)d,与单独应用Rapamycin组相比差异无统计学意义(P=0.948>0.05),但比单独用脾细胞组[(8.40±3.5d,P=0.013<0.05)]和未用药组有明显延长(P=0.015<0.05)。结论Rapamycin及联合应用供体脾细胞均可以延长移植心脏生存时间。Objective To observe the heart allografts survival time after administration of immunosuppressive agents Rapamycin or associated with donor splenoeytes on the time of operation. Methods A simplified heterotopie rat heart transplantation model was established and the Rapamycin (1 mg/kg) were vein injected or associated with donor splenocytes (1 × 10^9 cells/100 g). The grafts survival was observed by palpating over the abdominal wall every postoperation day. Results In using Rapamyein group, the average of the survival of heart allograft is( 12.3 ±3.9) days, had a significant difference compared with those of the no therapy group[ (8.5 ± 2.4)days, P = 0. 018 〈 0.05 ) 1. In using Raparnycin + donor splenoeytes group, the average survival of heart allografts is( 12.4 ± 3.7 ) days, had no signifieant difference with those of using Raparnycin alone ( P =0.018〈05 ), and much longer than those of using donor splenoeytes grout) [ ( 8.4 ± 3.5 ) days, P = 0. 013 〈 0.05 ] and no therapy group ( P =0. 015 〈0.05). Conclusion The findings demonstrate that a single injection of Rapamyein or associated with donor splenoeytes in rat can induce stable h)ng - term acceptance of heart allografts.
