摘  要:从本实验室分离短小芽孢杆菌B-15能降解羽毛角蛋白生产角蛋白酶,本试验对该菌株的产酶条件进行了优化。结果表明,在羽毛发酵培养基中,B-15产酶的最佳辅助碳源为葡萄糖,最佳辅助氮源为酵母粉,最佳接种量为2%,培养基最佳初始pH值为7.0,产酶最佳温度为30℃,最佳培养时间为48h。在以上条件下培养,角蛋白酶活力可达2.03U/mL,是优化前的2.5倍。对B-15所产的角蛋白酶进行了粗酶酶学性质的初步研究,结果表明,该酶最佳反应温度为55℃;最佳反应pH值为7.0;pH5.5~8.0时酶活力较稳定,可达最大酶活力的70%以上。Mg2+对酶有激活作用,Fe3+对酶有抑制作用。羽毛被降解后,其残渣和发酵后的液体可以作为动物的蛋白饲料。A strain of feather degrading bacteria was isolated which was be named Bacillus pumilus B-15 ,which can produce keratinase on feather.This study optimized the fermentation condition of B-15.The results showed that:the optimum carbon source was glucose;the optimum nitrogen source was yeast extract powder;the optimum quantity of inoculation was 2 % ;the optimum original pH was 7.0;the optimum cultural temperature was 30 ℃ and the optimum cultural time was 48 h.Under these conditions, the keratinase activity was 2.03 U/mL which was two point five times of the previous.Studied the properties of Bacillus pumilus B-15 ,the results showed that:the optimum temperature was 55 ℃ ;the optimum original pH was 7.0;The keratinase was stabile at pH 5.5 - 8.0.The keratinase could be activated by Mg^2+ and inhibited by Fe^3+.The residue of feather can be animal' s proteic feeds.
