摘 要:目的探讨姜黄素在大鼠体内的代谢行为。方法选择健康雄性大鼠,灌胃给予姜黄素500mg/kg,连续收集0~16h累积尿样。取10ml原尿加入β-葡萄糖醛酸水解酶缓冲盐溶液,反应后得酶解后尿样。原尿液和酶解后尿样经提取、浓缩、离心后用液质联用法(LC/MS-MS)进行分析。结果检测到尿中存在姜黄素原形药及7种代谢产物,即二氢姜黄素、姜黄素的葡萄糖醛酸结合物、六氢姜黄素的葡萄糖醛酸结合物、四氢姜黄素、六氢姜黄素、四氢姜黄素的葡萄糖醛酸结合物、姜黄醇,其中二氢姜黄素为首次报道。结论姜黄素在大鼠体内主要先还原为姜黄素还原物,经进一步代谢与葡萄糖醛酸结合。Objective To investigate the metabolisms of curcumin in rats. Methods Urine samples from 0 to 16h were collected after a single dose of 500 mg/kg curcumin was administered to rats. Add β-glucuronidase buffer solution to 10ml urine, we got the hydrolyzed urine samples. The urine and hydrolyzed urine samples were extracted, concentrated and centrifugaled, and then analyzed with LCIMS-MS. Results Cureumin and its seven metabolites were identified in rats' urine, including dihydrocurcumin, glucuronidase conjugation of cureumin, glucuronidase conjugation of hexahydroeureumin, tetrahydrocureumin, hexahydrocureumin, glucuronidase conjugation of tetrahydrocureumin and Cinobufagin. The dihydrocureumin was reported firstly. Conclusion According to the experimental results in this study, the major metabolic pathway of curcumin in rats is that the curcumin is reduced, followed by conjugation with glucuronic acid.
