摘  要:目的:研究体外使用SHH、Ra、Fn组成的诱导体系诱导骨髓间充质细胞后的上清液对BMSCs向神经细胞分化的作用。方法:从健康志愿者髂骨处抽取骨髓4mL,体外分离、培养、扩增后用400ng/LSHH、0.5μmolRa和5μmolFn组成的诱导体系进行诱导,取诱导后12,24h的诱导液上清分别作用于BMSCs。7d后使用倒置显微镜和免疫组织化学方法观察诱导前后细胞的变化。结果:诱导7d之后各组均有部分细胞胞体收缩、伸长突起,表现为神经细胞的形态。12h组同SHH+Ra+Fn组NSE、MAP-2阳性细胞比例差异无显著(P>0.05)。12h组分化率高于24h组(P<0.05)。结论:12h后诱导液上清可以有效地诱导BMSCs向神经细胞分化。Objective:To investigate the effect of supernatant after induction by sonic hedgehog,retinoie acids, forskolin inducible system on differentiation of human bone marrow stromal cells into neuron-like cells in vitro. Methods:BMSCs harvested from adult normal human marrow,then were passaged and purified in culture medium. BMSCs were passaged three times before experiment,then purified BMSCs were induced by sonic hedgehog (SHH),retinoie aeids(Ra) and Forskolinb(Fn). Supernatant after induction were harvested on 12,24 h then purified BMSCs were induced by supermatant after induction of 12 h, 24 h, 7 days later, BMSCs were detected with immunoeytoehemisty staining for miemtubule associated protein-2(MAP-2),neuron specific enolase (NSE). Results:After 7 days of induetion ,some BMSCs exhibited immunoreaetivity. MAP-2 and NSE positive cells existed in all groups ,while NSE,MAP- 2 and NSE positive cells increased markedly in BMSCs treated with supernatant after induction of 12 h. There were significant diferenees (P〈0. 05). Conelusion:Supernatant after induction of 12 h can effectively induce human BMSds to diferentiate into neural cells in vitro.
