摘 要:目的了解基质金属蛋白酶-2(MMP-2)在肝组织中的表达情况及与肝硬化患者肝功能损害程度的关系。方法采用免疫组织化学方法(SP法)观察了7例正常肝组织和23例肝炎后肝硬化患者肝组织中MMP-2表达的情况。结果在肝硬化患者肝组织中发现MMP-2主要在肝细胞、血管内皮细胞、肝窦内皮细胞及胆管内皮细胞的胞浆中高表达,表达情况与肝硬化Child-Pugh A、B、C分级相关,随着Child-Pugh分级及纤维化程度增高,MMP-2表达逐渐减少,A、B级患者肝组织中MMP-2表达最多,与对照组比较明显增高(P<0.01),A、B级与C级之间比较均有统计学意义(P<0.05)。结论肝硬化Child-Pugh分级不同,MMP-2表达发生了不同的变化,提示MMP-2在肝硬化形成中有重要作用。Objective To detect the expression of MMP-2 in hepatic tissue and to explore the relation between serverity of hepatic injury and the pathogenesis of fiberosis.Methods The expresion of MMP-2 were detected by immunohistochemistry and result was analyzed by picture quantitative analysis technique using immunohistological examination(SP method).The expression of MMP-2 were observed in the liver tissue in 7 normal subjects and 23 liver cirrhosis cases with a history of hepatitis.Results Immnoreactive MMP-2 was mainly expressed in the cytoplasm of hepatocyte,the blood vessel endothelium,hepatic sinusoids and biliary endothelial cells..Expression level in patients was correlated with the Child-Pugh classification.As the Child-Pugh grading increased,the MMP-2 expression level decreased.The expression in Child-Pugh class A and B were significantly higher than that in class C(P〈0.01).Conclusion The results suggests that MMP-2 plays an important role in in the formation of hepatic fibrosis.Different Child-Pugh class show different expression level in the hepatic fibrosis.
