关节腔注射5%碳酸氢钠对TMJ间接损伤后MMP-3的拮抗作用研究

摘  要:目的:对家兔TMJ(Temporomandibular joint,TMJ)间接性损伤后于其关节腔内注射5%碳酸氢钠,研究碱性试剂对细胞外基质的作用。方法:应用关节病理损伤指数、关节软骨含量和关节软骨MMP-3(matrixmetalloproteinases,MMPs)免疫荧光染色等观察指标来检测实验组和对照组动物。结果:4周及8周实验组动物软骨含量均高于对照组,两者比较差异有显著性(P<0.05);4周及8周对照组的关节病理损伤指数均高于实验组,有统计学差异(P<0.05);4周及8周的关节软骨MMP-3免疫荧光染色积分实验组均低于对照组,差异有显著性(P<0.05)。结论:应用碳酸氢钠可显著提高TMJ间接损伤的关节软骨含量,降低MMP-3表达,减轻关节创伤性炎症。Objective: To study the degradation of alkaline reagents for extracellular matrix through injecting 5% sodium bicarbonate into the joint cavities after indirect injury of temporomandibular joints in rabbits. Methods: The animals of the experimental group and the control group were detected by observation criteria such as the injury index of articular pathology, the contents of articular cartilage and the articular cartilage MMP-3 ( matrixmetalloproteinases, MMPs) immunity fluorescence dyeing. Results. The animal cartilaginous contents of the experimental group at the fourth and the eighth weeks were higher than those of the control group and there was difference between the two groups ( P 〈 0.05 ). The injury index of articular pathology of the control group at the fourth and the eighth weeks was higher than that of the experimental group and there was difference between the two groups ( P 〈 0.05 ). The articular cartilage MMP-3 immunity fluorescence dyeing of the integral experimental group at the fourth and the eighth weeks was lower than that of the control group and there was difference between the two groups ( P 〈 0.05 ). Conclusion: Using sodium bicarbonate may obviously enhance the articular cartilaginous contents of TMJ indirect injury, reduce the MMP-3 expression and relieve the joint traumatic inflammation.
