摘 要:采用RT-PCR分析填饲前后朗德鹅11种组织PPAR基因的表达差异性,结果显示,除腹脂外,PPAR-α在其他10种组织中都有表达,其中较高表达于心脏、肝脏、肾脏、十二指肠和胸肌;中等程度表达于肌胃、全脑、腿肌;较低表达于脾脏和肺脏。填饲后发现该基因在肾脏中依然高表达,除肺脏中表达量升高外,在其他组织中表达量均下降。PPAR-γ较高表达于肝脏、脾脏、肾脏、十二指肠和腹脂,较低表达于其他组织。填饲后发现该基因在心脏、脾脏、肺脏、肾脏、肌胃中表达量升高,在肝脏、十二指肠、全脑、胸肌、腿肌中表达量基本保持不变,在腹脂中表达量降低。结果表明PPARs的表达具有组织特异性,填饲前后的表达量变化反映PPARs基因影响鹅肥肝的形成。The present study was designed to investigate the expression characterization of PPAR genes before and after overfeeding in geese. Quantitative RT-PCR was developed to analyze the expression characterization of PPAR-a and PPAR-r gene in heart,liver,spleen,lung,kidney,stomaeh,intestine,brain,ehest musele,leg muscle and adipose tissues. The results showed that the PPAR-a expression was high in heart,liver,kidney,intestlne and chest muscle,medium in stomach,brain and leg muscle,low in spleen and liver. But expression was not detected in adipose. After overfeeding,the expression was remain high in kldney,inereased in lung,and reduced in other tissues. PPAR-y expression was high in liver,spleen,kldney, intestine and adipose,lowe in other tissues. After overfeeding,the expressions were increased in heart,spleen,hung,kidney and stomach,were kept invariable in liver,intestine,brain, chest and leg muscle,was reduced in adipose. The results showed that tissue expression of PPARs were tissue-specific and reflected of PPARs function infatty liver formation.
