摘　　要：目的:探讨表没食子儿茶素没食子酸酯[(-)-epi-gallocatechin-3 gallate,EGCG]抑制人鼻咽癌CNE-2Z细胞系生长增殖的机制,寻找鼻咽癌治疗的新靶点.方法:用EGCG处理人鼻咽癌CNE-2Z细胞,应用MTT试验观察不同浓度的EGCG对癌细胞生长增殖的影响;流式细胞仪检测用药前后细胞周期的变化;用逆转录聚合酶链反应分析ras相关区域家族1A(RASSF1A)mRNA表达情况.结果:从CNE-2Z细胞生长曲线判定EGCG对细胞生长均有抑制作用;流式细胞分析结果显示,G0/G1期细胞增加,S期细胞减少;经EGCG处理后,细胞中均有RASSF1AmRNA表达,且表达随药物浓度增高而增强.结论:EGCG对体外培养的鼻咽癌细胞生长增殖的抑制作用,可能是由于其能使因甲基化而失活的RASSF1A基因再转录,诱导该基因重新表达的结果.AIM： To explore the effect of （ - ）-epigallocatechin-3 gallate（EGCG） on the growth of CNE-2Z cell line and find new target of nasopharyngeal carcinoma （ NPC ） therapy. METHODS： CNE-2Z cells were treated by EGCG at different concentrations. MTT was used to detect the effect of EGCG on the growth of CNE-2Z cells. Cell cycle redistribution was analyzed by flow cytometry. The expression of RASSFI A mRNA was detected by RT-PCR. RESULTS： EGCG, at any does, suppressed the growth of CNE-2Z cells. No expression of RASSFI A mRNA was found in CNE-2Z cells without EGCG treatment. Flow eytometry showed that proportion of cells in G0/G1 phase was significantly increased, while those in S phase were reduced in CNE-2Z cells treated with EGCG. EGCG induced RASSF1A expression in CNE2Z cells in a dose-dependent manner. CONCLUSION： EGCG can inhibit the growth of tumor cells by reactivating RASSFIA genes silenced by promoter methylation in CNE-2Z cells in vitro.
