摘  要:目的检测缝隙连接蛋白43(Connexin43,Cx43)在大鼠急性局灶性脑缺血模型心室肌中的表达,以探讨其与脑缺血后心律失常的关系。方法用线栓法建立大鼠右侧局灶性脑缺血模型,检测心电,观察心肌细胞电镜下的亚显微结构,用Western blot的方法,检测心室肌中Cx43表达的改变。结果电镜结果显示脑缺血后,心肌组织闰盘内桥粒和缝隙连接结构及分布改变。Western blot结果显示,缺血后2 h心室肌中Cx43的表达明显降低,分布紊乱。缺血后24 h,1 w心室肌中Cx43的表达随时间延长而增加,且均高于正常对照组。结论大鼠右侧局灶性脑缺血所引起的心律失常可能与心室肌Cx43蛋白表达和分布异常有关。Objective To investigate the expression of ventricular Connexin43 (Cx43) in acute focal cerebral ischemia rats and to probe the relationship between the expression of ventricular connexin43 and the arrhythmia caused by acute focal cerebral ischemia. Methods The acute cerebral ischemia rats were induced by occluding right middle cerebral artery. The electrocardiogram (ECG) of the rats was moni- tored. The ultrastructure of the rat myocardium was examined. The ventricular tissues were taken 2 h, 24 h,and 1 week after cerebral ischemia. The expression of Cx43 of the ventricular tissue was detected by Western blot. Results Changes of bridge corpuscles in intercalated discs and gap junctions in myocardial tissues of the ischemia rats were observed by electronic microscope. Decreased amounts of Cx43 were found in the ventricle 2 h after cerebral ischemia compared with the control group. The expression of Cx43 increased in the ventiScle 24 h and 1 w after cerebral ischemia and the increasing was in time-depended manner. Conclusion Abnormal expression of Cx43 protein in the Ventricular after cerebral ischemia may he related with the arrhythmia resulted from acute focal cerebral ischemia.
