摘　　要：目的:在蛋白水平及基因水平检测视网膜母细胞瘤细胞株血管内皮生长因子受体VEGFR3的表达。方法:通过S-P免疫组化、RT-PCR、载体构建及基因测序等方法,研究VEGFR3在Rb50上的表达。结果:S-P免疫组化法呈阳性结果,RT-PCR扩增出313bp强表达条带,与载体pGEX-6P-3正确连接,经基因测序与VEGFR3基因序列一致。结论:视网膜母细胞瘤Rb50细胞株在基因水平及蛋白水平均有VEGFR3的表达,为研究VEGF-C/VEGFR3信息通路对视网膜母细胞瘤生长、浸润的影响打下了基础。Objectlve：To study the expression of proteins and DNA of VEGFR3 in retinoblastoma cell line Rb50. Methods： To examine the expression of proteins and DNA of VEGFR3 in RbS0 through S -P immunohistochemistry, RT -PCR and insert the VEGFR3 gene into PMD18 -T vector and sequence it . Results： The proteins of VEGFR3 in Rb50 cells was positively detected by S - P immunohistochemistry, and RNA of VEGFR3 was amplified by RT - PCR whose products was about 313bp against the marker in the AGE,the PMD18 -T vector was properly joined by the VEGFR3 gene which was in completely correspondence with VEGFR3 sequence in gene bank. Conclusion： The DNA of VEGFR3 is expressed and its proteins can be detected in the membrane of retinoblastoma cell line Rb50. This found could serve as the basis for the research about correlations between the growth and infiltrating of retinoblastoma and the signal of VEGF - C/VEGFR3.
