摘 要:目的:观察不同浓度α-生育酚(α-tocopherol,α-T)对链脲菌素(Streptozotocin,STZ)引起的胰岛细胞活力、胰岛素分泌及抗氧化能力变化的影响。方法:用体外单层细胞培养的方法培养1~3d Wistar大鼠的胰岛细胞分为4组,经分组处理后采用MTT、放射免疫等方法检测各组胰岛细胞活力、胰岛素分泌量、总抗氧化能力(T-AOC)、超氧化物歧化酶(SOD)活性、一氧化氮合酶(NOS)活性、丙二醛(MDA)及NO2-/NO3-含量的变化。结果:STZ可诱导胰岛细胞活力、T-AOC、SOD活性明显减弱;胰岛素分泌量显著降低,从(186.78±6.78)降至(64.59±4.32);MDA及NO2-/NO3-含量升高(P<0.05);而NOS活性无明显改变(P>0.05)。α-T能有效抑制STZ引起的上述指标的变化(NOS活性除外),该效应具有一定的剂量依赖性。结论:α-T能够改善STZ对大鼠胰岛细胞的损伤,该效应可能与增强胰岛细胞清除自由基的能力有关,而与NOS活性的变化关系不大。Objective:To investigate the inhibition effects of different concentrations of α-tocopherol on pancreatic islet cell viability,insulin secretion,anti-oxidation competence induced by streptozotocin. Methods:Pancreatic islet cells from Wistar rats aged 1 to 3 clays were cultured in monolayer in vitro, and divided into 4 groups.Then,cell viability,insulin secretion,T-AOC, SOD activity,MDA levcl,NOS activity and NO2-/NO3- production were measured by differrent detection methods. Results: Streptozotocin significantly decreased cell viability,T-AOC activity,SOD activity and insulin release levcl(P〈0.05),while increased MDA level,NO2-/NO3-production (P〈0.05).These effects were blocked by α-tocopherol.The inhibition effects of α-tocopherol were shown in a dose-dependent fashion.Streptozotocin induced an indistinct change of NOS activity(P〉0.05),that could not be blocked by α-tocopherol. Conclusion: α-tocopherol can attenuate the damage of rats pancreatic islet cells induced by strcptozotocin,the mechanisms of which may be that α-tocopherol can reinforce the competence of scavenging free radicals of pancreatic islet cells and not related to NOS activity change.
