长春花碱3和24h处理TK6细胞TK基因突变试验比较研究

摘　　要：[目的]比较用长春花碱对人淋巴母细胞TK6处理3h和24h的诱变性。[方法]用长春花碱对TK6细胞分别进行3h和24h染毒处理,用微孔板法进行TK基因突变试验。计算TK6细胞的接种效率(PE)、相对存活率RS、相对悬浮增值率RSG、总的相对增长率RTG、突变频率MF。[结果]在3h处理条件下,随长春花碱浓度的增加,RS、RTG和RSG等均降低,中、高剂量水平已表现出明显的细胞毒性,但各剂量水平的MF与溶剂对照比较差异均有统计学意义(P﹤0.05);在24h处理条件下,其MF随VBL浓度的增加而增加,并有一定的剂量-反应关系。[结论]较长时间处理对于长春花碱等细胞周期依赖性的毒物是必要的。[Objective ] To compare the induction of VBL with 3h and 24h treatment in human lymphoblastoid cells TK6. [Methods] TK gene mutation assay was performed by microwell plate method with human lymphoblastoid cells TK6 after exposing to vinblastin for 3h and 24h respectively. [Results] The results of the study showed that there was no significant difference between the mutation frequency of the VBL-3h-treatment groups and the control group, while significant difference existed tween those of the VBL-24h-treatment and the control group. [ Conclusion ] The results indicate that longer exposure may be needed for vinblastin and other cell cycle dependent chemicals.
